Key Enzyme Classes

1. Thermostable DNA Polymerases (e.g., Taq, Pfu): Essential for the Polymerase Chain Reaction (PCR). These enzymes can survive the 95°C denaturation temperatures required to unzip DNA strands, allowing for exponential amplification of specific sequences.

2. Ligases: These enzymes act as "molecular glue," repairing breaks in the sugar-phosphate backbone of DNA. They are critical for "cloning," where a target gene is inserted into a plasmid vector.

3. Nucleases: These include restriction enzymes that recognize specific palindromic sequences of DNA to make precise cuts, and RNases which are often removed or inhibited during RNA extraction to prevent sample degradation.

I just wrapped up Week 2 of Break Ground: The Thomas Manor Diaries, and even though I’m posting a little late, the momentum hasn’t stopped.

Last week was all about securing the foundation—not just for the construction side of things, but spiritually and financially, too. I reviewed my credit reports, began organizing my financial documents, and even received the Phase 1 construction budget from my contractor (a major win! 🙌🏾).

📖 The scripture that grounded me in Week 2 was:

“Commit to the Lord whatever you do, and He will establish your plans.” – Proverbs 16:3

This verse reminded me that while planning is essential, it’s surrender and trust that really allow our steps to be established.
